Doktorská práce pojednává o bakterii Escherichia coli v potravinách. Práce se zabývá její charakterizací pomocí fenotypizačních a genotypizačních metod. Bakterie Escherichia coli se mohou vyskytovat v různých druzích potravin, zejména živočišného původu. Rezistence k antimikrobiálním látkám je v současnosti celosvětovým problémem, jelikož je zaznamenán vysoký nárůst rezistentních bakterií člověka a zvířat. Ke zjištění prevalence rezistentních kmenů E. coli v potravinách bylo provedeno testování na citlivost k vybraným antibiotikům diskovou difúzní metodou. Kromě toho byla také zjišťována přítomnost genů rezistence pomocí polymerázové řetězové reakce (PCR), zejména na beta-laktamová antibiotika. Jelikož se E. coli řadí mezi podmíněně patogenní mikroorganizmy, bylo dalším cílem zjišťování přítomnosti vybraných faktorů virulence pomocí klasické a multiplex PCR. Pro určení patogenního potenciálu izolovaných kmenů přispělo také zařazení kmenů do fylogenetických skupin metodou triplex PCR. Součástí charakterizace bylo taktéž stanovení schopnosti produkce bakteriocinů s následnou genotypizací pro určení přesného typu kolicinu nebo mikrocinu. V neposlední řadě byla zjišťována schopnost kmenů E. coli tvořit biofilm spektrofotometrickým měřením absorbance v mikrotitračních destičkách.
Získané výsledky přináší nové poznatky o antibiotické rezistenci a virulenci kmenů E. coli izolovaných z potravin a přispívají k hlubšímu poznání vlastností této bakterie. Na základě získaných výsledků byl zhodnocen význam a potencionální nebezpečí přítomnosti E. coli v potravinách a prostředí.

The doctoral thesis focuses on bacterium Escherichia coli in food. The PhD thesis deals with the characterization using phenotypic and genotypic methods. Escherichia coli is found in various kinds of food, especially in food of animal origin. Currently, resistance to antimicrobial agents is a worldwide problem because of a large increasing of resistant bacteria of humans and animals. To determine the prevalence of resistant E. coli strains from foodstuffs, antibiotics sensitivity testing to selected antibiotics by discs diffusion method has been carried out. Furthermore, it has been tested the presence of resistance genes by the PCR method, especially to beta-lactam antibiotics. Since E. coli belongs among potentially pathogenic microorganisms, another objective of characterization was to determined selected virulence factors using classical and multiplex PCR. Classification of strains into phylogenetic groups by triplex PCR also contributed to determine the pathogenic potential of isolates. Another part of characterization was to found out the production of bacteriocins with subsequent genotyping by PCR to determine the exact type of colicin or microcin. Finally, it was determined biofilm formation ability by spectrofotometrically measuring of absorbance in microtiter plates. All these results bring new insights about virulence and antibiotic resistance in E. coli strains isolated from food and contribute to a deeper understanding about physiological characteristics of this bacterium. Based on the obtained results was evaluated importance and potential dangers of the occurrence of E. coli in food and environment.
