Předložená disertační práce byla zaměřena na zhodnocení biologické rozložitelnosti polyvinylalkoholu (PVA) za aerobních, anaerobních a zejména denitrifikačních podmínek, a to s využitím čistírenských mikrobiálních kultur. Denitrifikační podmínky byly zabezpečeny odstraněním kyslíku z degradačních suspenzí a přídavkem dusičnanů, které sloužily přítomným mikroorganismům jako akceptory elektronů při anaerobní respiraci. Studium biodegradace PVA za uvedených podmínek bylo zvoleno z důvodu zcela nedostatečného prozkoumání této oblasti. Disertační práce se zabývala studiem kinetiky mikrobiální degradace PVA za denitrifikačních podmínek, která byla porovnána s rychlostí biologického rozkladu za anaerobních i aerobních podmínek. Biodegradace polyvinylalkoholu byla hodnocena na základě úbytku jeho koncentrace v průběhu pokusů, dále byly sledovány změny v koncentraci dusičnanů a také hodnoty redox potenciálů kalových suspenzí. Součástí disertační práce byl i podrobný výzkum směřující k poznání klíčových mikroorganismů v mikrobiálním společenstvu rozkládající polyvinylalkohol za denitrifikačních podmínek a řada experimentů byla věnována možnostem isolace degradačních bakterií.

This doctoral thesis was focused on the evaluation of the polyvinyl alcohol (PVA) biodegradation under aerobic, anaerobic and especially denitrifying conditions, with the use of sludge inoculums originated from the common wastewater treatment plant. Denitrifying conditions were ensured by an oxygen absence and by nitrates addition at the beginning of the tests; the nitrates served as electron acceptors for anoxic respiration of present microorganisms. The main reason for this doctoral study was very poor scientific knowledge concerning PVA biodegradation under denitrifying conditions. Biodegradation kinetic under denitrifying conditions was compared with the processes proceeding under aerobic and anaerobic conditions.
The degree of PVA biodegradation in the tests was evaluated by the direct spectrophotometric measurement of PVA concentration, determination
of dissolved organic carbon and changes in nitrate ions concentration. The values of redox potentials were checked in all the tests. The important part of the doctoral thesis was focused on the observation of composition of the microbial community degrading polyvinyl alcohol under denitrifying conditions and on the many attempts to isolation of key bacterial degrader.
